U bolnici u New Yorku prvi put je transplantiran svinjski bubreg u čovjeka bez izazivanja trenutnog odbacivanja imunološkog sistema primaoca, što je potencijalno veliki napredak koji bi na kraju mogao pomoći u ublažavanju nedostatka ljudskih organa za transplantaciju, piše The New York Times.
Postupak napravljen u bolnici “NYU Langone Health” u New Yorku uključivao je upotrebu svinje čiji su geni bili promijenjeni tako da njezina tkiva više nisu sadržala molekulu za koju je poznato da izaziva skoro trenutno odbacivanje.
Primalac je bila pacijentkinja kojoj je mozak klinički mrtav, a sa znacima disfunkcije bubrega, čija je obitelj pristala na eksperiment prije nego što ju je trebalo skinuti sa uređaja za održavanje života, rekli su istraživači za Reuters.
Tri dana je novi bubreg bio pričvršćen za njene krvne sudove i održavan izvan tijela, dajući istraživačima pristup do njega.
Rezultati ispitivanja funkcije transplantiranog bubrega “izgledali su prilično normalno”, rekao je kirurg za transplantaciju dr. Robert Montgomery, koji je vodio studiju.
Bubreg je napravio “količinu urina koju biste očekivali” iz transplantiranog ljudskog bubrega, rekao je on, a nema dokaza o snažnom, ranom odbacivanju viđenom kada se nemodifikovani svinjski bubrezi transplantiraju u primate koji nisu ljudi.

Nenormalan nivo kreatinina u primaocu – pokazatelj slabe funkcije bubrega – vratio se u normalu nakon transplantacije, rekao je Montgomery.
U SAD-u skoro 107.000 osoba trenutno čeka na transplantaciju organa, uključujući više od 90.000 koji čekaju bubreg, prema Ujedinjenoj mreži za doniranje organa. Prosječno vrijeme čekanja na bubreg je tri do pet godina.
Istraživači su desetljećima radili na mogućnosti korištenja životinjskih organa za transplantaciju, ali su bili u nedoumici kako spriječiti ljudsko tijelo da ih odmah odbaci.
Montgomeryijev tim je teoretizirao kako bi izbacivanje svinjskog gena za ugljikohidrat koji izaziva odbacivanje – molekul šećera ili glikan, nazvan alfa-gal – spriječilo problem.
Eksperiment transplantacije bubrega u New Yorku trebalo bi otvoriti put za ispitivanja kod pacijenata sa završnim stadijem bubrežne insuficijencije, vjerovatno u narednih godinu ili dvije, rekao je Montgomeri, koji je i sam primalac transplantiranog srca. Ta ispitivanja mogla bi testirati pristup kao kratkoročno rješenje za kritično bolesne pacijente dok ne postane dostupan ljudski bubreg ili kao trajni transplantat.

Postupak napravljen u bolnici “NYU Langone Health” u New Yorku uključivao je upotrebu svinje čiji su geni bili promijenjeni tako da njezina tkiva više nisu sadržala molekulu za koju je poznato da izaziva skoro trenutno odbacivanje.
Primalac je bila pacijentkinja kojoj je mozak klinički mrtav, a sa znacima disfunkcije bubrega, čija je obitelj pristala na eksperiment prije nego što ju je trebalo skinuti sa uređaja za održavanje života, rekli su istraživači za Reuters.
Tri dana je novi bubreg bio pričvršćen za njene krvne sudove i održavan izvan tijela, dajući istraživačima pristup do njega.
Rezultati ispitivanja funkcije transplantiranog bubrega “izgledali su prilično normalno”, rekao je kirurg za transplantaciju dr. Robert Montgomery, koji je vodio studiju.
Bubreg je napravio “količinu urina koju biste očekivali” iz transplantiranog ljudskog bubrega, rekao je on, a nema dokaza o snažnom, ranom odbacivanju viđenom kada se nemodifikovani svinjski bubrezi transplantiraju u primate koji nisu ljudi.

Nenormalan nivo kreatinina u primaocu – pokazatelj slabe funkcije bubrega – vratio se u normalu nakon transplantacije, rekao je Montgomery.
U SAD-u skoro 107.000 osoba trenutno čeka na transplantaciju organa, uključujući više od 90.000 koji čekaju bubreg, prema Ujedinjenoj mreži za doniranje organa. Prosječno vrijeme čekanja na bubreg je tri do pet godina.
Istraživači su desetljećima radili na mogućnosti korištenja životinjskih organa za transplantaciju, ali su bili u nedoumici kako spriječiti ljudsko tijelo da ih odmah odbaci.
Montgomeryijev tim je teoretizirao kako bi izbacivanje svinjskog gena za ugljikohidrat koji izaziva odbacivanje – molekul šećera ili glikan, nazvan alfa-gal – spriječilo problem.
Eksperiment transplantacije bubrega u New Yorku trebalo bi otvoriti put za ispitivanja kod pacijenata sa završnim stadijem bubrežne insuficijencije, vjerovatno u narednih godinu ili dvije, rekao je Montgomeri, koji je i sam primalac transplantiranog srca. Ta ispitivanja mogla bi testirati pristup kao kratkoročno rješenje za kritično bolesne pacijente dok ne postane dostupan ljudski bubreg ili kao trajni transplantat.
