Onaj koji ljubimca nije imao nema pojma koji je to osjećaj duboke tuge kada vas psić ili maca napuste. A moraju. Jer - vrijeme...
Psi žive znatno kraće od ljudi, pa se neizbježan trenutak rastanka približi poput treptaja, već za kojih desetak, petnaest godina.
Kažu oni koji se razumiju kako je jedna naša godina jednaka sedam psećih, e pa taj bi se termin mogao ubrzo promijeniti. U tijeku je završna faza procesa proizvodnje medikamenata kojima je zadatak produžiti život psima. Doznajemo kako bi se na tržištu taj čaroban lijek mogao naći već početkom 2026. godine.
Lijek još uvijek nema osmišljen tržišni naziv, već je poznat po svojoj tehnološkoj oznaci LOY-001, na njemu radi i razvija ga američka tvrtka Loyal, smještena u San Franciscu.
Ukratko su pojasnili "u čemu je kvaka", kazavši kako je lijek stvoren za velike, odrasle pse, koji inače žive kraće od malenih, radi na način da se bavi inzulinoikim faktorom rasta (IGF), hormonom koji utječe na rast, a samim tim i ubrzavanje starenja, a uobičajeno je visok upravo kod većih vrsta pasa. Dokazali su znanstveno, naime, kako su psi s nižom razinom inzulina manje osjetljiviji i da kvalitetnije žive od drugih.
U igri su vitamini i minerali
LOY-001 će svako tri do šest mjeseci veterinari injektirati psima. Neće biti plasiran na tržište u obliku tablete. Tako će se pod kontrolom držati korištenje, premda, vele znanstvenici, "straha nema", jer su dosadašnja istraživanja pokazala kako nuspojava nema, osim blagih i privremenih gastrotegoba...
Loyal čeka da njihov LOY-001 odobri Američka agencija za hranu i lijekove (U.S. Food and Drug Administration, FDA), pa su pred njima duga klinička ispitivanja...

- Ne možemo točno znati o čemu se radi, jer nema objavljenih podataka. Nisu vitamini i minerali u igri, to je sigurno. No, inhibitor hormona rasta kod velikih pasmina - ima smisla. Izvršna direktorica (CEO) Loyala kaže da jedan od lijekova ima sličan mehanizam kao rapamicin (koji se već istražuje za tu svrhu), koji inhibira aktivnost TORC1. TORC1, ili "Target of Rapamycin Complex 1", je proteinski kompleks koji igra ključnu ulogu u regulaciji rasta i metabolizma u mnogim eukariotskim organizmima, uključujući i ljude. TORC1 je dio većeg signalnog puta poznatog kao mTOR (mammalian Target of Rapamycin) put, koji je od vitalnog značaja za kontrolu mnogih bioloških procesa, uključujući sintezu proteina, autofagiju (proces razgradnje i recikliranja staničnih komponenti), te odgovor na stres i dostupnost hranjivih tvari, objašnjava nam dr. sc. Šuran.
Rapamicin je lijek koji selektivno inhibira mTOR put, posebno ciljajući TORC1. Veže se za protein FKBP12, a ovaj kompleks zatim interagira s TORC1, inhibirajući njegovu aktivnost. Kroz ovu inhibiciju, rapamicin može usporiti ili zaustaviti rast stanica, što ga čini korisnim u tretmanu određenih vrsta raka, u prevenciji odbacivanja transplantiranih organa, pa eto i u istraživanju starenja i dugovječnosti.
Put do ‘pilule za vječnost‘
- Djelovanje rapamicina i njegov utjecaj na TORC1 put je predmet intenzivnih istraživanja, jer razumijevanje ovih mehanizama može pružiti uvide u tretmane raznih bolesti, uključujući rak, neurodegenerativne bolesti, i možda čak pridonijeti razvoju terapija usmjerenih na produženje zdravog života - tako će profesorica.
No, pitanje je o čemu se točno radi. Na "mreži svih mreža" sve je dostupno pa smo pronašli i patent tvrtke Loyal, čija je direktorica i sama dijelom tima koji ga je izumio.

- Patentom je zaštićena primjena "terapijski učinkovite količine nukleinske kiseline koja kodira ciljani protein ili funkcionalni fragment ili varijantu istog, ili terapijski učinkovite količine modificiranog proteina koji sadrži ciljani protein ili funkcionalni fragment ili varijantu istog", što znači da su ti lijekovi, osim inhibitora TORC1, i neka varijanta nukleinskih kiselina koje su od razgradnje zaštitili i povećali biodostupnost uklapanjem u liposome i/ili lipidne nanočestice - objasnila nam je farmakologinja.
Još se neki inhibitori mTOR istražuju: poput everolimusa (RAD001) koji je sličan rapamicinu, u biti je derivat rapamicina koji također inhibira mTOR put, posebno ciljajući TORC1. Koristi se u tretmanu određenih vrsta raka, kao što su bubrežni karcinom i neuroendokrini tumori, kao i u prevenciji odbacivanja transplantiranih organa. Potom temsirolimus (CCI-779) koji je još jedan derivat rapamicina, inhibira mTOR put i primjenjuje se u liječenju naprednog bubrežnog karcinoma - veli nam Šuran.
I tada dolazimo do onog što nam svima postaje jasno. Do "pilule za vječnost"...
- I vjerojatno će, kad dobiju dovoljno podataka na psima, a možda i odobrenje FDA, nastaviti na ljudima. Ako to već ne rade - kazala nam je znanstvenica.

Onaj koji ljubimca nije imao nema pojma koji je to osjećaj duboke tuge kada vas psić ili maca napuste. A moraju. Jer - vrijeme...
Psi žive znatno kraće od ljudi, pa se neizbježan trenutak rastanka približi poput treptaja, već za kojih desetak, petnaest godina.
Kažu oni koji se razumiju kako je jedna naša godina jednaka sedam psećih, e pa taj bi se termin mogao ubrzo promijeniti. U tijeku je završna faza procesa proizvodnje medikamenata kojima je zadatak produžiti život psima. Doznajemo kako bi se na tržištu taj čaroban lijek mogao naći već početkom 2026. godine.
Lijek još uvijek nema osmišljen tržišni naziv, već je poznat po svojoj tehnološkoj oznaci LOY-001, na njemu radi i razvija ga američka tvrtka Loyal, smještena u San Franciscu.
Ukratko su pojasnili "u čemu je kvaka", kazavši kako je lijek stvoren za velike, odrasle pse, koji inače žive kraće od malenih, radi na način da se bavi inzulinoikim faktorom rasta (IGF), hormonom koji utječe na rast, a samim tim i ubrzavanje starenja, a uobičajeno je visok upravo kod većih vrsta pasa. Dokazali su znanstveno, naime, kako su psi s nižom razinom inzulina manje osjetljiviji i da kvalitetnije žive od drugih.
U igri su vitamini i minerali
LOY-001 će svako tri do šest mjeseci veterinari injektirati psima. Neće biti plasiran na tržište u obliku tablete. Tako će se pod kontrolom držati korištenje, premda, vele znanstvenici, "straha nema", jer su dosadašnja istraživanja pokazala kako nuspojava nema, osim blagih i privremenih gastrotegoba...
Loyal čeka da njihov LOY-001 odobri Američka agencija za hranu i lijekove (U.S. Food and Drug Administration, FDA), pa su pred njima duga klinička ispitivanja...
O svemu smo popričali s dr. sc. Jelenom Šuran, direktoricom biotech start-upa Apiotix Technologies, bivšom profesoricom veterinarske farmakologije na zagrebačkom Veterinarskom fakultetu.

- Ne možemo točno znati o čemu se radi, jer nema objavljenih podataka. Nisu vitamini i minerali u igri, to je sigurno. No, inhibitor hormona rasta kod velikih pasmina - ima smisla. Izvršna direktorica (CEO) Loyala kaže da jedan od lijekova ima sličan mehanizam kao rapamicin (koji se već istražuje za tu svrhu), koji inhibira aktivnost TORC1. TORC1, ili "Target of Rapamycin Complex 1", je proteinski kompleks koji igra ključnu ulogu u regulaciji rasta i metabolizma u mnogim eukariotskim organizmima, uključujući i ljude. TORC1 je dio većeg signalnog puta poznatog kao mTOR (mammalian Target of Rapamycin) put, koji je od vitalnog značaja za kontrolu mnogih bioloških procesa, uključujući sintezu proteina, autofagiju (proces razgradnje i recikliranja staničnih komponenti), te odgovor na stres i dostupnost hranjivih tvari, objašnjava nam dr. sc. Šuran.
Rapamicin je lijek koji selektivno inhibira mTOR put, posebno ciljajući TORC1. Veže se za protein FKBP12, a ovaj kompleks zatim interagira s TORC1, inhibirajući njegovu aktivnost. Kroz ovu inhibiciju, rapamicin može usporiti ili zaustaviti rast stanica, što ga čini korisnim u tretmanu određenih vrsta raka, u prevenciji odbacivanja transplantiranih organa, pa eto i u istraživanju starenja i dugovječnosti.
Put do ‘pilule za vječnost‘
- Djelovanje rapamicina i njegov utjecaj na TORC1 put je predmet intenzivnih istraživanja, jer razumijevanje ovih mehanizama može pružiti uvide u tretmane raznih bolesti, uključujući rak, neurodegenerativne bolesti, i možda čak pridonijeti razvoju terapija usmjerenih na produženje zdravog života - tako će profesorica.
No, pitanje je o čemu se točno radi. Na "mreži svih mreža" sve je dostupno pa smo pronašli i patent tvrtke Loyal, čija je direktorica i sama dijelom tima koji ga je izumio.

- Patentom je zaštićena primjena "terapijski učinkovite količine nukleinske kiseline koja kodira ciljani protein ili funkcionalni fragment ili varijantu istog, ili terapijski učinkovite količine modificiranog proteina koji sadrži ciljani protein ili funkcionalni fragment ili varijantu istog", što znači da su ti lijekovi, osim inhibitora TORC1, i neka varijanta nukleinskih kiselina koje su od razgradnje zaštitili i povećali biodostupnost uklapanjem u liposome i/ili lipidne nanočestice - objasnila nam je farmakologinja.
Još se neki inhibitori mTOR istražuju: poput everolimusa (RAD001) koji je sličan rapamicinu, u biti je derivat rapamicina koji također inhibira mTOR put, posebno ciljajući TORC1. Koristi se u tretmanu određenih vrsta raka, kao što su bubrežni karcinom i neuroendokrini tumori, kao i u prevenciji odbacivanja transplantiranih organa. Potom temsirolimus (CCI-779) koji je još jedan derivat rapamicina, inhibira mTOR put i primjenjuje se u liječenju naprednog bubrežnog karcinoma - veli nam Šuran.
I tada dolazimo do onog što nam svima postaje jasno. Do "pilule za vječnost"...
- I vjerojatno će, kad dobiju dovoljno podataka na psima, a možda i odobrenje FDA, nastaviti na ljudima. Ako to već ne rade - kazala nam je znanstvenica.
