Ljudi koji boluju od Alzheimerove bolesti, vjerojatno nisu svoju memoriju "izgubili" zauvijek nego su im pohranjene informacije teže dostupne, utvrdio je tim kineskih znanstvenika predvođenih nobelovcem Susumijem Tonegavom.
Oni su predložili i nove tarapije kojima bi se u budućnosti mogle liječiti pogubne posljedice demencije.
Istraživači su proveli pokuse na miševima kojima su pomoću plavog svjetla stimulirali određene dijelove mozga. Životinje su se pomoći tih podražaja sjetile informacija koje im više nisu bile dostupne.
U pokusu su koristili genetski modificirane miševe kod kojih su se razvili simptomi slični onima koje ima više od 30 milijuna ljudi širom svijeta koji pate od Alzheimerove bolesti.
Miševi su smješteni u kutiju po čijem je podu prolazila električna struja tako da su životinje u nogama osjetile neugodan i opasan eletrični šok. Kada su nakon 24 sata miševi ponovno stavljeni u kutiju, oni koji nisu imali Alzheimerovu bolest od straha su ostale na mjestu, jer su predvidjele da će uslijediti strujni udar.
Stimuliranje engramskih stanica
Kada su u kutiju stavljeni miševi s Alzheimerovom bolešću, takvo oprezno ponašanje nije se dogodilo, iz čega znanstvenici zaključuju da se ne sjećaju svoj prvog neugodnog iskustva.
No kada su te miševe osvijetlili plavim svjetlom, čime su htjeli stimulirati njihove engramske stanice povezane s memorijom, miševi su se ponašali kao da su se sjetili bolnog iskustva.
Isti rezultat dobili su i kada su tijekom električne stimulacije miševe premjestili u drugu kutiju, što je dokaz da je sjećanje sačuvano i da se aktiviralo.
Kada su ispitali mozak ovih miševa, otkrili su da miševi s Alzheimerovom bolesti imaju manje "vodiča" kroz koje nastaju sinaptičke veze između moždanih stanica.
Svjetlosnom stimulacijom uspjeli su povećati broj "vodiča" na broj sličan onome kod zdravih miševa, što je rezultiralo njihovim "normalnim" ponašanjem u očekivanju strujnog udara.
Budući da ljudi i miševi imaju slične osnovne principe djelovanja memorije, zaključci upućuju da Alzheimerova bolest ne uništava sjećanje, nego ga samo čini nedostupnim.
"Memorija bi, dakle, barem u prvim fazama bolesti, moga biti sačuvana u mozgu, ali je problem u tome kako je ponovno dozvati. To znači da možda lijek postoji", pojasnio je Tonegava.
Je li rješenje u optogenetici?
Za provođenje optičke stimulacije moždanih stanica, tehnikom koja je nazvana optogenetika – moraju se u moždane stanice najprije ugraditi posebni geni koji omogućuju da neuroni postanu osjetljivi na plavo svjetlo, a potom slijedi stimuliranje određenih dijelova mozga plavim svjetlom.
Optogenetski pristup se već u prošlosti koristio u liječenju duševnih bolesti kao što su depresija i posttraumatski stresni poremećaj.
No prije nego što se ta nova tehnologija počne koristiti za liječenje bolesnika s Alzheimerovom bolesti trebat će obaviti cijeli niz dodatnih istraživanja, kazao je Tonegava.

Ljudi koji boluju od Alzheimerove bolesti, vjerojatno nisu svoju memoriju "izgubili" zauvijek nego su im pohranjene informacije teže dostupne, utvrdio je tim kineskih znanstvenika predvođenih nobelovcem Susumijem Tonegavom.
Oni su predložili i nove tarapije kojima bi se u budućnosti mogle liječiti pogubne posljedice demencije.
Istraživači su proveli pokuse na miševima kojima su pomoću plavog svjetla stimulirali određene dijelove mozga. Životinje su se pomoći tih podražaja sjetile informacija koje im više nisu bile dostupne.
U pokusu su koristili genetski modificirane miševe kod kojih su se razvili simptomi slični onima koje ima više od 30 milijuna ljudi širom svijeta koji pate od Alzheimerove bolesti.
Miševi su smješteni u kutiju po čijem je podu prolazila električna struja tako da su životinje u nogama osjetile neugodan i opasan eletrični šok. Kada su nakon 24 sata miševi ponovno stavljeni u kutiju, oni koji nisu imali Alzheimerovu bolest od straha su ostale na mjestu, jer su predvidjele da će uslijediti strujni udar.
Stimuliranje engramskih stanica
Kada su u kutiju stavljeni miševi s Alzheimerovom bolešću, takvo oprezno ponašanje nije se dogodilo, iz čega znanstvenici zaključuju da se ne sjećaju svoj prvog neugodnog iskustva.
No kada su te miševe osvijetlili plavim svjetlom, čime su htjeli stimulirati njihove engramske stanice povezane s memorijom, miševi su se ponašali kao da su se sjetili bolnog iskustva.
Isti rezultat dobili su i kada su tijekom električne stimulacije miševe premjestili u drugu kutiju, što je dokaz da je sjećanje sačuvano i da se aktiviralo.
Kada su ispitali mozak ovih miševa, otkrili su da miševi s Alzheimerovom bolesti imaju manje "vodiča" kroz koje nastaju sinaptičke veze između moždanih stanica.
Svjetlosnom stimulacijom uspjeli su povećati broj "vodiča" na broj sličan onome kod zdravih miševa, što je rezultiralo njihovim "normalnim" ponašanjem u očekivanju strujnog udara.
Budući da ljudi i miševi imaju slične osnovne principe djelovanja memorije, zaključci upućuju da Alzheimerova bolest ne uništava sjećanje, nego ga samo čini nedostupnim.
"Memorija bi, dakle, barem u prvim fazama bolesti, moga biti sačuvana u mozgu, ali je problem u tome kako je ponovno dozvati. To znači da možda lijek postoji", pojasnio je Tonegava.
Je li rješenje u optogenetici?
Za provođenje optičke stimulacije moždanih stanica, tehnikom koja je nazvana optogenetika – moraju se u moždane stanice najprije ugraditi posebni geni koji omogućuju da neuroni postanu osjetljivi na plavo svjetlo, a potom slijedi stimuliranje određenih dijelova mozga plavim svjetlom.
Optogenetski pristup se već u prošlosti koristio u liječenju duševnih bolesti kao što su depresija i posttraumatski stresni poremećaj.
No prije nego što se ta nova tehnologija počne koristiti za liječenje bolesnika s Alzheimerovom bolesti trebat će obaviti cijeli niz dodatnih istraživanja, kazao je Tonegava.
