Rezultati nedavne studije, objavljeni u veljači u medicinskom časopisu Acta Neuropathologica, pokazuju da se prvi znaci Alzheimerove bolesti mogu pojaviti u očima.

"Oko je prozor u mozak", kaže oftalmologinja Christine Greer, direktorica medicinskog obrazovanja na Institutu za neurodegenerativne bolesti u Boca Ratonu u američkoj sveznoj državi Floridi. "Možete vidjeti izravno u živčani sustav promatrajući stražnji dio oka, prema optičkom živcu i mrežnici", dodala je ona.

Cilj istraživanja, kako javlja CNN, a prenosi Jutarnji list, bio je pokušati utvrditi kako analiza oka može pomoći u dijagnosticiranju Alzheimerove bolesti prije pojave samih simptoma. Kod oboljelih osoba, upozoravaju stručnjaci, bolest je već uvelike uznapredovala do trenutka kada budu pogođeni pamćenje i ponašanje osobe.

"Alzheimerova bolest počinje se razvijati u mozgu desetljećima prije prvih simptoma gubitka pamćenja", ističe Richard Isaacson, neurolog specijaliziran za Alzheimerovu bolest s Instituta za neurodegenerativne bolesti.

Promjene u mrežnici i mozgu

Ukoliko bi liječnici uspjeli identificirati bolest u njezinim najranijim fazama, ljudi bi mogli odabrati zdrav način života i kontrolirati svoje "promjenjive čimbenike rizika, poput visokog krvnog tlaka, visokog kolesterola i dijabetesa", rekao je Isaacson koji, inače, nije sudjelovao uprovođenju istraživanja.

Kako bi doznali koliko rano možemo uočiti znakove kognitivnog propadanja, u sklopu nedavnog istraživanja znanstvenici su analizirali donirano tkivo mrežnice i mozgova 86 osoba koje su prolazile kroz različite stupnjeve mentalnog pada.

"Naša je studija prva koja pruža dubinske analize proteinskih profila i molekularnih, staničnih i strukturnih učinaka Alzheimerove bolesti na ljudsku mrežnicu i kako oni korespondiraju s promjenama u mozgu i kognitivnim funkcijama", rekla je koautorica studije Maya Koronyo-Hamaoui, profesorica neurokirurgije i biomedicinskih znanosti u neprofitnom medicinskom centru Cedars-Sinai u Los Angelesu.

"Te promjene u mrežnici povezane su s promjenama u dijelovima mozga koji se nazivaju entorhinalni i temporalni korteks, središte za pamćenje, navigaciju i percepciju vremena", pojašnjava Koronyo-Hamaoui.

Istraživači su tijekom provođenja studije prikupljali uzorke retinalnog i moždanog tkiva tijekom 14 godina 86 ljudskih donora tkiva s Alzheimerovom bolešću i blagim kognitivnim oštećenjem što je, orema autorima, najveća skupina retinalnih uzoraka ikada proučavana.

Potom su usporedili uzorke donatora s normalnom kognitivnom funkcijom s onima koji su imali blaga kognitivna oštećenja i onima s poodmaklim stadijima Alzheimerove bolesti. Studija je otkrila značajno povećanje beta-amiloida - ključnog markera Alzheimerove bolesti - kod osoba s Alzheimerovom bolešću i ranim kognitivnim padom.

Ranije dijagnosticiranje

Mikroglijalne stanice propale su za 80 posto kod onih s kognitivnim problemima, pokazalo je istraživanje. Radi se o stanicama odgovornim za obnovu i održavanje drugih stanica, što uključuje čišćenje beta-amiloida iz mozga i mrežnice.

"Pronađeni su (također) i markeri upale, koji bi mogli biti jednako važan pokazatelj napredovanja bolesti", rekao je Isaacson. "Nalazi su bili očiti i kod osoba bez ili s minimalnim kognitivnim simptomima, što ukazuje na to da bi ovi novi očni testovi mogli biti vrlo korisni pri postavljanju rane dijagnoze".

Istraživači su otkrili i veći broj imunoloških stanica koje čvrsto okružuju amiloidne beta plakove, kao i druge stanice odgovorne za upale i smrt stanica i tkiva. Atrofija tkiva i upala u stanicama na krajnjoj periferiji mrežnice najviše ukazuju na to kakav je kognitivni status, pokazalo je istraživanje.

"Ova otkrića bi na kraju mogla dovesti do razvoja slikovnih tehnika koje bi nam omogućile ranije i točnije dijagnosticiranje Alzheimerove bolesti... te praćenje njezinog napredovanja neinvazivnim putem promatranjem kroz oko", zaključuje Isaacson, prenosi Jutarnji list.

"Oko je prozor u mozak", kaže oftalmologinja Christine Greer, direktorica medicinskog obrazovanja na Institutu za neurodegenerativne bolesti u Boca Ratonu u američkoj sveznoj državi Floridi. "Možete vidjeti izravno u živčani sustav promatrajući stražnji dio oka, prema optičkom živcu i mrežnici", dodala je ona.

Cilj istraživanja, kako javlja CNN, a prenosi Jutarnji list, bio je pokušati utvrditi kako analiza oka može pomoći u dijagnosticiranju Alzheimerove bolesti prije pojave samih simptoma. Kod oboljelih osoba, upozoravaju stručnjaci, bolest je već uvelike uznapredovala do trenutka kada budu pogođeni pamćenje i ponašanje osobe.

"Alzheimerova bolest počinje se razvijati u mozgu desetljećima prije prvih simptoma gubitka pamćenja", ističe Richard Isaacson, neurolog specijaliziran za Alzheimerovu bolest s Instituta za neurodegenerativne bolesti.

Promjene u mrežnici i mozgu

Ukoliko bi liječnici uspjeli identificirati bolest u njezinim najranijim fazama, ljudi bi mogli odabrati zdrav način života i kontrolirati svoje "promjenjive čimbenike rizika, poput visokog krvnog tlaka, visokog kolesterola i dijabetesa", rekao je Isaacson koji, inače, nije sudjelovao uprovođenju istraživanja.

Kako bi doznali koliko rano možemo uočiti znakove kognitivnog propadanja, u sklopu nedavnog istraživanja znanstvenici su analizirali donirano tkivo mrežnice i mozgova 86 osoba koje su prolazile kroz različite stupnjeve mentalnog pada.

"Naša je studija prva koja pruža dubinske analize proteinskih profila i molekularnih, staničnih i strukturnih učinaka Alzheimerove bolesti na ljudsku mrežnicu i kako oni korespondiraju s promjenama u mozgu i kognitivnim funkcijama", rekla je koautorica studije Maya Koronyo-Hamaoui, profesorica neurokirurgije i biomedicinskih znanosti u neprofitnom medicinskom centru Cedars-Sinai u Los Angelesu.

"Te promjene u mrežnici povezane su s promjenama u dijelovima mozga koji se nazivaju entorhinalni i temporalni korteks, središte za pamćenje, navigaciju i percepciju vremena", pojašnjava Koronyo-Hamaoui.

Istraživači su tijekom provođenja studije prikupljali uzorke retinalnog i moždanog tkiva tijekom 14 godina 86 ljudskih donora tkiva s Alzheimerovom bolešću i blagim kognitivnim oštećenjem što je, orema autorima, najveća skupina retinalnih uzoraka ikada proučavana.

Potom su usporedili uzorke donatora s normalnom kognitivnom funkcijom s onima koji su imali blaga kognitivna oštećenja i onima s poodmaklim stadijima Alzheimerove bolesti. Studija je otkrila značajno povećanje beta-amiloida - ključnog markera Alzheimerove bolesti - kod osoba s Alzheimerovom bolešću i ranim kognitivnim padom.

Ranije dijagnosticiranje

Mikroglijalne stanice propale su za 80 posto kod onih s kognitivnim problemima, pokazalo je istraživanje. Radi se o stanicama odgovornim za obnovu i održavanje drugih stanica, što uključuje čišćenje beta-amiloida iz mozga i mrežnice.

"Pronađeni su (također) i markeri upale, koji bi mogli biti jednako važan pokazatelj napredovanja bolesti", rekao je Isaacson. "Nalazi su bili očiti i kod osoba bez ili s minimalnim kognitivnim simptomima, što ukazuje na to da bi ovi novi očni testovi mogli biti vrlo korisni pri postavljanju rane dijagnoze".

Istraživači su otkrili i veći broj imunoloških stanica koje čvrsto okružuju amiloidne beta plakove, kao i druge stanice odgovorne za upale i smrt stanica i tkiva. Atrofija tkiva i upala u stanicama na krajnjoj periferiji mrežnice najviše ukazuju na to kakav je kognitivni status, pokazalo je istraživanje.

"Ova otkrića bi na kraju mogla dovesti do razvoja slikovnih tehnika koje bi nam omogućile ranije i točnije dijagnosticiranje Alzheimerove bolesti... te praćenje njezinog napredovanja neinvazivnim putem promatranjem kroz oko", zaključuje Isaacson, prenosi Jutarnji list.
